Technical AFLP (Amplified Fragment Length Polymorphism) was used to determine the level of genetic diversity of wild and cultivated species of the genus Polianthes as only been linked based on their morphological characteristics. 11 individuals were analyzed, 9 wild species of P. montana, P. howardii, P. platiphylla, P. geminiflora var. Clivicola, P. zapopanensis, P. geminiflora var. Pueblenensis, P. pringleii, P. tuberosa var. Double and P. tuberosa var. Simple, along with 2 external controls Prochnyanthes and Manfreda. In the analysis bands by combination of primers was found that the combination Mse + CAT / Eco + ACT had the highest total number of ands. However; It was the combination with the lowest percentage of polymorphism (85.71%), unlike the other two combinations (Mse + CAG / Eco + ACA) and (Mse + CAG / Eco + ACC) where the total number of bands was 95 and 91 respectively, but both combinations polymorphism showed a level of 100%. The product of the level of polymorphism found in individuals could be separated into a cluster dendrogram with average similarity of all individuals of 0.603 with the formation of 3 major groups, Group I consists of individuals of P. montana, P. zapopanensis, P. pueblensis, P. tuberosa var. Double and P. tuberosa var. Simple; Group II: formed by individuals of the species P. howardii, P. platyphylla, P. geminiflora var. Clivicola, and Group III: formed by individuals of P. platyphylla and P. geminiflora platyphylla var. Clivicola and the individual's gender Prochnyantes. Indicating a wide diversity within the genre as most wildlife is demonstrating the utility of molecular markers such as AFLP to establish phylogenetic relationships between organisms and estimate the variation within populations of Polianthes. | |
